I can't quite recall as it's been quite a while since I've played the game, but it's shortly after the Pendleton twins mission where you find Emily in the brothel, possibly after the Lady Boyle mission.

Short term? I'm not sure. Just doing it once won't have a physical toll, providing that you don't accidentally kill yourself or whatever. Long term use will most likely have some health problems, just like every other drug.

Also had an idea for the sequel. You know how Overseers show up in weird ass places? Brigmore manor and the like? Keep that in mind. The most interesting part of the plot to me was actually at the very beginning, when you give the Empress the letter. She announces that the other kingdoms are going to blockade Dunwall. Now consider this. SPOILERS.
Three regime changes in six months (Empress-> Lord Regent-> Havelock-> Emily.
A plague still burning the city.
Political domestic disarray. The people and guards don't know who to follow.
The other kingdoms see their chance to strike. War comes to Dunwall. You play an Overseer, who has been visited by the Outsider (which you struggle to keep secret). You are tasked with traveling throughout the other kingdoms, seeking aid to help Dunwall survive, and maybe, just maybe find a way to stop the war.
Just a thought.

I'd like to see the sequel set in some other island/city of the Empire. Perhaps the bodies of the plague victims from Dunwall drifted to some other city and carried the plague there. Perhaps instead of rats being a major motif this time, it could be crows feasting on the bodies of the deceased. How cool would it be to summon a swarm of murderous crows instead of rats? Makes sense too, since 'Corvo' comes from the Latin 'corvus', meaning 'crow'.

Just a thought.

Arkane Studios confirmed Dishonored as a new IP, so there will be another game, but they haven't revealed what the game is going to be about or when it's going to go into development.

There's been a lot of speculation that you'll play as a grown up Emily in the second game, mainly because she often took great interest in Corvo's work, asking him to teach her swordfighting & various martial arts techniques, and there's also a point in the game where she finds a rune and keeps it under her pillow in the hopes it would bring her good luck, but eventually gives it to Corvo claiming it gave her nightmares instead (You can see her talking in her sleep during one of these nightmares if you visit her room at the Hound Pits Pub, and it's heavily implied she sees the Outsider in her dreams, suggesting she may also be "gifted" like Daud & Corvo) . Nothing is set in stone yet, but it could be a cool concept, seeing as in 2 of the 3 endings during the epilogue, Corvo serves as her bodyguard, thwarting assassination attempts several times until she eventually grows up and Corvo passes away, after which point she'll obviiously have to fend for herself since her bodyguard will no longer be there. EIther way, I'm interested to see what their concept will be, DIshonored was probably my favorite game of 2012, there just aren't enough AAA Steampunk games =/

For the sequel, I don't think Corvo will be our protagonist. In the DLC, the Outsider tells Daud, "Did you know there are only eight like you in the world? WHo bear my mark?" That means that there are eight possible protagonists for eight possible games. Whatever happens, I can't wait.

Most of the negative effects are purely psychological. It's been linked to a number of psychoses and lower intelligence, but whether marijuana use is a cause or an effect of these is still very much up for debate. It does cause temporary serotonin or dopamine (I forget which) depletion, which is what causes the lack of motivation associated with stoners, however this is temporary and goes away quickly (This is often called a weed hangover, though not physically painful like an alcohol hangover, I personally find it much harder to work with). With continuous use, the depletion can result in full-blown depression. And finally, there's the fact that you're typically burning it. All combustion produces a number of known carcinogens. However, people don't typically smoke weed nearly as often as they smoke tobacco, so the likelihood of developing a cancer is significantly lower with marijuana than tobacco. Really though, most of this is speculative as very little research has been done with marijuana use due to legal restrictions. There is little conclusive research to suggest that marijuana is generally "bad" for you, but it's still a drug. All drugs have their pros and cons, whether it be aspirin or heroin.

TLDR: There isn't much about weed that is super bad for you, but like any other drug it does carry some relatively minor side effects.

Just like cigarettes, you're still inhaling smoke. Cigarettes have a bunch of other poisonous shit though, but smoke is smoke nonetheless. And people can do some pretty stupid things when they're high, so there's that. And I don't know if it actually makes you stupid or not, but every stoner I knew in high school was pretty damn stupid.
