Long story short, humans wiped themselves out with a nuclear holocaust and all the mystical creatures, candy people, and talking animals are all actually mutants born from the radiation.

Actually in one of the newest episodes "Simon & Marcy" you actually see what is possibly the first candy person, I horribly mutated blob of pink with a face.

Perhaps she ages differently than most others?   
Although it isn't clearly stated as a 100% rule of her aging, her age appears to be associated with the mass of her body, which is why she became younger when she was blown up, and then older when they combined her body with parts of the other candy people to make up for the missing candy in her body...   
With this logic, it is plausible for her to be that old

I agree, especially when they hint several times in the series of Marcy and PB having some sort of past. I.e the Door Lord episode.

I suppose pinning age to a sometimes homicidal humanoid made out candy is kind of silly. I just figured ancestor is easier to explain but right now all we can go off is speculation, I actually like how the writers keep certain aspects of the story vague, keeps things interesting when you can theorize about the backstory.
